چکیده

Introduction: Alzheimer’s disease (AD) affects 35 million people worldwide (Dartigues, 2009). AD research using MRI has focused on early changes in medial temporal and cortical gray matter. Although caudate atrophy is more typical of motor disorders such as Parkinson’s disease (Nakamura et al. 2001), beta-amyloid and tau pathology accumulates in the caudate in AD (Braak and Braak, 1990). Here we mapped 3D patterns of caudate atrophy in a large cohort of AD, mild cognitive impairment (MCI), and matched elderly control subjects. We tested whether caudate atrophy is associated with: (1) clinical scores (MMSE, CDR) and their decline over a 1-year follow-up period; (2) conversion from MCI to AD; (3) immediate and delayed Logical Memory scores from the Wechsler Memory Scale; (4) CSF A, taup-tau levels; and (5) age, sex, ApoE genotype, and body mass index. We used cumulative distribution functions and false discovery rate theory (FDR) to rank the relative strengths of these associations.

MRI Biomarkers and Modifiable Health Factors of Aging and Dementia

MRI research examining structural brain atrophy in Alzheimer’s disease (AD) generally focuses on medial temporal and cortical structures, but amyloid and tau deposits also accumulate in the caudate. Here we mapped the 3D profile of caudate atrophy using a surface mapping approach in subjects with AD and mild cognitive impairment (MCI) to identify potential clinical and pathological correlates. ...

Multimodal Magnetic Resonance Imaging in Alzheimer’s Disease Patients at Prodromal Stage

One objective of modern neuroimaging is to identify markers that can aid in diagnosis, monitor disease progression, and impact long-term drug analysis. In this study, physiopathological modifications in seven subcortical structures of patients with mild cognitive impairment (MCI) due to Alzheimer's disease (AD) were characterized by simultaneously measuring quantitative magnetic resonance param...

Effects of Baseline CSF α-Synuclein on Regional Brain Atrophy Rates in Healthy Elders, Mild Cognitive Impairment and Alzheimer’s Disease

BACKGROUND Cerebrospinal fluid (CSF) α-synuclein is reduced in synucleinopathies, including dementia with Lewy bodies, and some studies have found increased CSF α-synuclein in Alzheimer's disease (AD). No study has explored effects of CSF α-synuclein on brain atrophy. Here we tested if baseline CSF α-synuclein affects brain atrophy rates and if these effects vary across brain regions, and acros...

Comparing the Diagnostic Value of Four Dementia Tests in the Amnestic and Healthy Elderly

Objective: This study aimed to compare the diagnostic value of 4 questionnaires for the diagnosis of neurocognitive disorders (NCDs) in the elderly. Methodology: In this project, people older than 60 years who lived in Tehran were investigated. A total of 99 literate persons were enrolled in the study, and 4 questionnaires of functional assessment staging tool (FAST), abbreviated mental test s...

Vertex-wise shape analysis of subcortical structures in Alzheimer’s disease

Background: Most MRI studies in Alzheimer’s disease (AD) have focused on cortical and hippocampal atrophy. This study, however, examines regional volumetric and shape abnormalities of subcortical and limbic structures in AD. Methods: Thirty-six patients with AD and 36 matched controls were included in the study. All subjects were right-handed, evaluated using standard clinical assessment scales...
